Phenylpropanolamine (PPA) is a sympathomimetic drug that is a vasoconstrictor and bronchodilator. In 2000, it was removed from the market due to causing stroke. Research on a relationship between PPA exposure and hair cell development has not yet been observed. The zebrafish (Danio rerio) is a useful model for studying ototoxic effects on hair cells because the lateral line system along the outside of their body is composed of hair cells. Ototoxic effects on the hair cells of the lateral line system can help determine ototoxic effects on the hair cells of the inner ear of mammals and humans. The lateral line system is a mechanosensory organ that senses water flow. When stimulated in zebrafish larvae an escape response is evoked. Using this knowledge, tests were done to stimulate lateral line and determine if there was a difference in the reaction times of larvae which developed in various concentrations of PPA ranging from 0.25 to 1.5 mM until 5 days post fertilization (dpf). Additionally, photos were taken and heart rate was observed after incubation in PPA and after over-night recovery. Reaction time results were determined to be inconclusive. However, heart rate results showed that there was a significant decrease in the heart rate of larvae treated with 0.50 and 0.75 mM PPA until 5dpf. After recovery, larvae treated with 0.25 and 0.50 mM PPA experienced a significant increase in heart rate which was not significantly different from the control. Differences in morphology were observed among concentration groups.

Wild and captive grey wolves (Canis lupus) not only live in different habitats, but they also have different necessary survival activities. These differences give rise to questions concerning exhibit familiarity and the correlation with activity levels. In the present study, activity budgets of male grey wolves from two different locations with differing levels of exhibit familiarity were observed. The results of this study show that there is a significant difference between the activity levels and exhibit use of habituated wolves at the Milwaukee County Zoo and the unhabituated wolves at the Menominee Park Zoo. It is also shown that there is no significant change in activity over time in either wolf pack.

This study examined primary succession on the basin of a catastrophically drained lake in northern Minnesota. The site, Bass Lake, suffered this drainage as a result of logging operations in 1925, which reduced the total area of the lake from approximately 2 km2 present day size of 1.05 km2 (260 ac.). I hypothesized that succession occurring on this newly exposed area would follow the relay floristics model of Clements. Specifically, I expected a change from shade-intolerant trees, such as Betula papyrifera (paper birch), to shade-tolerant trees, such as Abies balsamea (balsam fir). To test this hypothesis, I used data collected using the Point-Centered Quarter Method for trees found below the old shoreline between the years of 1978 and 2010. The results indicated that there was a transition from paper birch to balsam fir on the north side of the lake. However, paper birch remained as the most significant tree on the south side of the lake throughout the study’s duration with no such transition. As a result, it appears that the north side of Bass Lake follows the relay floristics, but this hypothesis must be rejected for the south side. Factors such as soil texture, water availability, canopy openings, and growing season may have caused this difference.

As a regulator of transcription, antitermination works to allow for RNA polymerase to read through termination signals and express genes found downstream of these termination signals. The Q protein of phage λ displays this antitermination phenotype in E.coli. It works to allow for the expression of promoter distal genes by letting RNAP transcribe through termination signals that block these genes. Q function overlays the phage late gene promoter P R ’ and requires the presence of a DNA element called the Q-utilization (qut) site. The qut site must contain Q-binding sequences and a specific pause site for the Q protein to be able to modify RNAP for the antitermination phenotype. Mutations in the qut site (-13/- 15) have been shown to decrease antitermination efficiency by preventing Q protein binding. This research seeks to identify regions of the Q protein that interact with qut DNA, using a second site suppressor analysis to identify mutant Q genes that allow for antitermination phenotype on mutant (-13/- 15) qut sites.

This study was undertaken to validate the use of noninvasive techniques for measuring androgen hormone metabolites in the Eastern Bluebird (Sialia sialis). Typically researchers have used blood plasma to measure direct hormone levels within vertebrates; however, there are drawbacks to this method. The initial stress of handling can negatively influence results due to stress hormone interference. Also, the small size of the subject limits your sample volumes, so it is difficult to get a full view of the changes in hormone levels over time. Noninvasive methods are desirable because samples are easily obtained, do not harm the subject, and provide a better view of the pattern of hormone levels over time. This technique must be validated for every species because of variations in specific metabolic pathways. In this study, we used a GnRH challenge to analyze the responsiveness of the HPG axis in adult male Eastern Bluebirds through both testosterone levels in their blood and T-metabolite concentrations in their droppings. Results determined that both blood plasma testosterone and excrement T-metabolites rose in response to the GnRH challenge in such a way that we were able to determine that the noninvasive method of measurement is valid for Eastern Bluebirds.

The rotifer Brachionus plicatilis is a halophilic invertebrate often used for research and feeding young fish. One concern in this work is to provide young fish with energy-rich lipids. Past research suggests that neutral lipids serve a primarily structural role in rotifers, while other research suggests that neutral lipids are broken down for energy during starvation of rotifers, but these ideas are not mutually exclusive. In this research, Nile Red staining technique was used to quantify neutral lipids during starvation coupled with liquid chromatography mass spectrometry (LCMS) to identify specific lipids present during periods of starvation. Results demonstrate that lipids are used for energy during periods of starvation up to the 48–hour point and that there is a change in fatty acid composition over longer periods of starvation. Thus, lipids appear to be an important biochemical in the storage of energy in rotifers.

For a long time, Iliotibial Band Syndrome (ITBS), a common injury seen in athletes of all different backgrounds, was believed to be due to friction between the iliotibial band (ITB) and the lateral femoral epicondyle of the femur. However, Fairclough et al. 2006 recently proposed a new compression model that looks at the compression of a highly innervated fatty tissue layer beneath the ITB, instead of the pain stemming from the ITB itself. This new model has led researchers to rethink previous beliefs and conduct more research to better understand the syndrome. Under further examination, MRI’s back up Fairclough et al. by revealing the presence of inflammation beneath the ITB in a layer of highly innervated fatty tissue that becomes tightly compressed during 30° flexion, an angle that can be reached by an activity as simple as climbing a flight of stairs. Further research has been performed to reevaluate two precursors that have been found to be associated with ITBS in the past, increased peak hip adduction and increased peak internal knee rotation, to determine whether it would be possible for them to lead to the new compression syndrome. Due to the ability of both of these precursors to pull the ITB tighter than it normally would be, it was concluded that they could increase the pressure the ITB places on the highly innervated fatty tissue layer, resulting in the compression syndrome.

Cell death caused by cardiac infarction events commonly result in the development of heart disease. The identification and classification of genes in these cell death pathways improves medical professionals understanding of these mechanisms and helps in the development of gene therapy techniques which can be used to impede the onset of heart disease. The goal of this research was to identify novel genes in cell death pathways. We amplified cDNA for 10 genes which were screened for high death potential. This amplified cDNA was then added into an inducible vector and inserted into an adenovirus. This adenovirus was then introduced to cells with the doxycycline (DOX) inducer and examined for cell death. We expected to see cells that had virus applied without DOX to be healthy and living, and cells with virus and DOX to have significant cell death from the induced gene. Instead, we observed substantial cell death in both cases of virus applied to cells and healthy, living control cells. We concluded that the induced gene could not be responsible for this death and rather viral toxicity caused the increased death. Further testing these viruses, with consideration of recent literature, could demonstrate if these genes are active in a cell death pathway.

Because Escherichia coli and other coliform bacteria pose a risk to public health through
waterborne illnesses, it is necessary to investigate the impact of well depth and septic tank proximity on bacterial contamination in rural private wells. I hypothesized that coliform contamination in wells correlates to shallow well depth and proximity to a household septic tank. To test this hypothesis, I sampled 182 wells at rental properties in Vilas County, a rural county in north central WI. My results showed that well depth and septic tank proximity had no significant influence on total coliform acteria or E. coli contamination. Additional studies with larger sample sizes are required to determine the effect of well depth, septic tank distance, and other well construction factors on coliform contamination in groundwater wells.

Understanding forage species’ energy density is important for mapping energy flow in an ecosystem. Bioenergetic models are used to determine which species play important roles and how changes may influence the ecosystem in which they reside. In the Northwest Atlantic ecosystem, energy flow is largely controlled by forage species. The current study aims to 1) compare two forage species’ energy content and 2) find a size measurement which can be used to predict energy content. Using a Soxhlet apparatus and muffle furnace, the lipid and protein values of two forage species, Alewife and Silver Hake, were collected. Alewife are 3-4 times as lipid rich as Silver Hake, depending on the size measurement. Dry weight measurements can be used to predict energy in Alewife, however, Silver Hake did not have a size measurement which could be used to confidently predict energy content. Why species vary in which measurements can be used to predict energy and how to effectively use the energy data that can be collected is a direction for future research.

Transcription termination is an important regulatory component within the Escherichia coli (E. coli) genome. Two proteins are known to influence transcriptional termination in E. coli: Rho and NusA. Rho only functions to induce transcription termination, while NusA is involved in both the enhancement and inhibition of termination. This study aimed to determine whether the two proteins influence each other's functions through direct or indirect interactions. Reciprocal yeast two hybrid (Y2H) analysis using vectors expressing Rho and NusA as fusion with the protein domain of the Gal4 transcriptional factor indicated direct interactions between the two proteins. These results will allow us to further study the mechanistic roles of each protein in transcription termination, as well as the process of gene expression throughout the E.coli genome.
